Colonization Dynamics of Multidrug-Resistant Klebsiella pneumoniae Are Dictated by Microbiota-Cluster Group Behavior over Individual Antibiotic Susceptibility: A Metataxonomic Analysis

Gastrointestinal carriage of multidrug-resistant (MDR) bacteria is one the main risk factors for developing serious, difficult-to-treat infections. Given that there currently no all-round solution to eliminate colonization with MDR bacteria, it particularly important understand dynamic process aid development novel decolonization strategies. The aim our present study was perform metataxonomic analyses gut microbiota dynamics during an extended-spectrum β-lactamase (ESBL)- and carbapenemase-producing Klebsiella pneumoniae (ECKP) strain in mice; additionally, ascertain effects antibiotic administration (ampicillin, ceftazidime, ciprofloxacin) on establishment elimination ECKP intestinal colonization. We have found phyla Bacteroidetes Firmicutes were most dominant all treatment groups; however, more common groups treated antibiotics compared control group. Significant differences observed among different antibiotic-treated beta but not alpha diversity, implying difference relative abundance some bacterial community members. Bacteria from Lachnospiraceae family (including Agathobacter, Anaerostipes, Lachnoclostridium 11308, UCG-004, NK3A20 group 11318, NK4A136 11319, Roseburia, Tyzzerella) showed inverse relationship rate strain, whereas members Enterobacteriaceae shown a correlational relationship. Our results suggest composition microbial plays primary role MDR-colonization rate, susceptibility individual strains affects this lesser extent. Distinct families associated into clusters, collecting taxonomically close species produce survival benefits gut. These associations do develop at random, as they may be attributed presence specific metabolomic networks. A new concept should introduced designing future endeavors decolonization, supplemented by knowledge host identification clusters capable suppressing or enhancing invader species.
